I'd like to play off a USB and play it when I have downtime at work. I downloaded the game to my PC and copied it to my USB, but when I tried running the .exe on my work computer I got an error saying there was something wrong with the serial.

I can play the game no problems on my home PC from the USB. I also play the game with no issues on my Steam account too on my PC.

I understand though if it's not possible for anti-piracy reasons.

No it's not possible. You'll have to install the game using the game file installer on each computer you want to play.
