I suspect I know the answer to this already, but I figured I'd try for confirmation:

Does the hero on the Special Units (like Oleh Dir or Albert Kerscher) take the place of the first hero that would normally be awarded in the 100-300 kill range?

(Meaning that these units have to reach the 301-1000 kill range to start working on their second hero.)

You're right. You can only have three heroes, so the special hero takes the first slot. Then you can have two more which are awarded for the the same number of kills as the second and third heroes for a normal unit.
