Two key heroes will be Prudent and Consolidator, but Prudent is the absolutely most important one. It will ensure that the damaged actually strengthens the defensive capabilities and Consolidator with overstrenght gives more health to benefit from the ability. Granted, you do not want to heal such a unit. You leave it damaged and at one point the extra defense gets so high that the infantry is invulnerable to all but the most insanely powerful damage dealers like custom forts or Landkreuzers.
This gives you two more heroes to work with. No Surrender is useful, as it is possible run into trouble if the unit gets heavily suppressed, but not damaged and then hit by a powerful attack. Not available normally, but might by in modded campaigns, is No Retreat, serving a very similar role. If you can, run both. If not, the fourth hero can be either Zero Slots (or at least Reduced Slots), so the side effect of Consolidator is negated. Another option is Vigilant, if you plan to use any other infantry then Pioneers for this type of defensive unit, as normal Infantry does not have as good Close Defense value.

Clear choices are Readiness, Aggressive Counterattack, Low Profile or Evasive, and Resilient. These will make the tank very tough to kill and if you manage to get Survivor on it, it should shine in defensive conditions.

Depends if you want the unit to suppress the target so others can do the job, or destroy such target on its own, this will vary. It will also be different for artillery and planes:
Suppression, artillery: Double Attack, Shock Tactics, Zero Slots, Kloss or Habermann - use on rocket artillery
Destruction, artillery: Double Attack, Lethal Attack, Zero Slots, Kloss or Habermann - use on 21cm artillery, alternative late war is a Sturmtiger. Rocket artillery can work early, but will struggle with late war armor.
Suppression, aircraft: Double Move, Double Attack, Shock Tactics, Lightning Attack - use on strategic bombers
Destruction, aircraft: Double Move, Double Attack, Ignores Entrenchment, Lightning Attack - use on dive bombers, Rudel can replace a few of these heroes giving even more powerful options such as overstrenght
