In the last desperate attempt at winning the campaign the Orks used weaponized Vortex tech to open a rift into Warp on the planet surface, and a bunch of Chaos troopers and demons appeared wishing to settle a bill with the Space Wolves from a much earlier conflict. They flattened the Orks and then barely got flattened by Wolves and Imperial Guard themselves. It was a single battle, not an entire campaign, but should do as backstory.

Tyranids are my favorites and would love to see that army sooner than later, but I would be pretty damn happy with some generic (Ultra)marines that gave us some color scheme choices for more than one chapter.

Reading about Sanctus reach, maybe the DLC will be Cadian armoured imperial guard?

Personally Necron and Sisters of Battle.

1 - Chaos Armies (just a "Dark" repaint of the in game Marines so should be easiest)
2 - Eldar (just a cool race over-all) ;|
3 - Imperial Guard (billions strong, who can beat that)

The ability to use other chapters, of course.
Just make the option to use Imperial Fists, or Black Templars. I'd replay the entire game

Other than that, I'd say heavy bolters are lacking, graviton weapons would be a blast, and I'd love to roll out a Thunderfire cannon

I would vote for Sisters of Battle and the Grey Knights for good guys... Bad guys I would have to vote for Eldar and Necron...

Or any other third party other than Imperium. Its really bad if there is two faction of same side but only orks as their enemies.
UNLESS you guys make world Armageddon campaing, with new ork units as well. There is whole books of it
Butt still multiplayer wise, it needs to be third party from Imperium and Orks.

Astra Militarum and some generic Space Marines, so you can make your own chapters, with a bit of skinning.

1. Eldar, obviously. From the good old times of Dreamforge's Rites of War we are waiting for it (Dawn(s) of War do not count due to casual and non-tabletop feeling).
2. Other Chapters to choose from.
3. Tyranids/Chaos - one of two of them... or both
